冬蟲夏草對乳癌誘發噬骨細胞分化的影響

提要
依衛生署統計(2009),在台灣乳癌的發病率佔女性惡性腫瘤的第一位,而死亡率
則佔第四位。乳癌細胞常轉移至骨組織,誘發噬骨細胞過度活化,而導致骨組織破壞。
在我們先前的研究中,發現保健食品中的冬蟲夏草,可以加強骨髓經放射或化學治療的
功能回復。本實驗發現冬蟲夏草新的功效,即抑制由乳癌細胞所誘發的噬骨細胞生成及
骨組織分解。本研究結果發現,人類乳癌細胞株(MDA-231)的條件培養液會促進血液
周邊單核球分化成噬骨細胞,且含酸性磷酸酶之細胞及骨片吸收面積比例亦會增加。人
類乳癌細胞所引起的上述效應會被冬蟲夏草所抑制,其機轉可能透過壓制RANKL 及
IL-6 的基因表現所造成。結論:實驗中證實冬蟲夏草可經由抑制噬骨細胞分化而減緩乳
癌細胞的骨吸收能力,顯示冬蟲夏草對於抑制乳癌轉移至骨組織為一有潛力的藥物。

Effects of Cordyceps sinensis on differentiation of
osteoclast induced by breast cancer

Abstract
According to Taiwan’s Department of Health statistics 2009, the morbidity of breast
cancer has been the first of women’s malignancies, and the fourth leading cause of cancer
death in Taiwanese women. Human breast cancer frequently metastasizes to bone tissues
associated with bone destruction as a consequence of the activation of osteoclasts (OC). We
have previously reported the potential of the health food supplement Cordyceps sinensis (CS)
as a remedy that can enhance the recovery of bone marrow function following radiotherapy or
chemotherapy. In this study, we identify a new function of CS as a remedy to prevent the
osteoclastogenesis and osteolytic bone invasion of a human breast cancer. The results
demonstrate that the condition medium of MDA-231 cells could drive the differentiation of
human peripheral blood monocytes (PBMC) into OC as demonstrated by the increase of
tartrate-resistant acid phosphatase (TRAP) staining and the percentages of bone resorption
area per bone slices. These effects were diminished by the addition of CS, which effects
could be contributed to the suppression of receptor for activation of NF-κB ligand (RANKL),
and IL-6. In summary, this study demonstrates that CS is a potential remedy to prevent the
metastatic potential of malignant breast cancer cells by interfering tumor-directed bone
resorption action through its effect on osteoclastogenesis.
